What is the function of the DTNA gene?
DTNA encodes for α-dystrobrevin, a component of the macromolecular dystrophin-glycoprotein complex that binds to dystrophin/utrophin and α-syntrophin. Absence of α-dystrobrevin causes muscular dystrophy in mice but variants in DTNA in humans have not been associated with muscle disease. DMD, the largest known human gene, provides instructions for making a protein called dystrophin. This protein is located primarily in muscles used for movement (skeletal muscles) and in heart (cardiac) muscle. Small amounts of dystrophin are present in nerve cells in the brain.In DMD, muscle is missing a key structural protein called dystrophin, making it more susceptible to injury. Eventually the body cannot keep up with repair and regeneration, leading to muscle loss (wasting) and weakness.A mutation in the donor splice junction at the end of dystrophin exon 62 was associated with human Duchenne Muscular Dystrophy. Dystrophin gene mutations are associated with duchenne muscular dystrophy. Duchenne Muscular Dystrophy (DMD) is caused by deletion, duplication or point mutation of the dystrophin gene.
